What Is Orforglipron (Foundayo)?

Foundayo is an FDA-approved oral prescription medicine from Eli Lilly. Its active ingredient, orforglipron, is a non-peptide, small-molecule GLP-1 receptor agonist. Boxed warning: risk of thyroid C-cell tumors. Orforglipron is not pharmacologically active in rats or mice and did not produce tumors in rodents. The human relevance of GLP-1 receptor-dependent rodent thyroid C-cell tumors has not been determined. Foundayo is contraindicated with a personal or family history of medullary thyroid carcinoma (MTC), MEN 2, or known serious hypersensitivity to orforglipron or its excipients. Mechanism of Action

Orforglipron binds to and activates the human GLP-1 receptor. GLP-1 is a physiological regulator of appetite and caloric intake. The label reports reduced food intake, likely mediated by decreased appetite, and delayed gastric emptying. With a reduced-calorie diet and increased physical activity, Foundayo is indicated to reduce excess body weight and maintain weight reduction long term in adults with obesity, or adults with overweight and at least one weight-related comorbid condition. Label §1.

Concomitant use with another GLP-1 receptor agonist is not recommended.
